The Tesco store in Truro will close for five days from Saturday.
Bosses said the Garras Wharf store would close for improvements from 7pm on Saturday to 8am on Friday, April 12.
During the closure the pharmacy will be open for prescriptions only.
A spokesman for Tesco said the store will benefit from a new pizza counter, a Click and Collect service, Costa Coffee, Rowe's pasty counter and will introduce scan as you shop.
There will also be a new look to the deli, meat and fish counter, fruit and veg department, bakery, clothing, phone shop, and pharmacy.
Customers previously told the store they wanted more space to shop, new fixtures and a fresher look.
Store manager Stuart Bourn said: "It is really important to us that we keep disruption to a minimum during this short closure.
"Our main priority is keeping our customers happy and we have listened to all feedback ahead of making these major improvements.
"My team and I would like to thank our customers in advance for their patience whilst we improve their store."
The nearest Tesco store is at Helston on Clodgey Lane, which is open from 6am Monday – 10pm Saturday and 10am-4pm on a Sunday or the St Austell store based on Daniels Lane which is open from 6am Monday – midnight Saturday and 10am-4pm on a Sunday.
